چکیده
Abstract Adipose-derived stem cell (ADSC) is one of the most widely used candidate for intervertebral disc (IVD) degeneration-related disease. However, poor survival and low differentiation efficacy in stressed host microenvironment limit therapeutic effects ADSC-based therapy. The preconditioning has been found effective to boost proliferation functioning cells varying pathological condition. Lithium a common anti-depression drug proved enhance functioning. In this study, using LiCl on cellular behavior ADSC was investigated, specially degenerative IVD-like Method toxicity rat assessed by detecting lactate dehydrogenase (LDH) production after treatment with concentration lithium chloride (LiCl). proliferative capacity determined Ki67 expression relative number ADSC. Then, preconditioned challenged And viability as well nucleus pulpous (NP) evaluated major marker extracellular matrix (ECM) deposit. were an IVD degeneration model, NP morphology ECM content assessed. Results A range 1–10 mmol/L applied following since higher causes death (about 40%). similar between groups control group preconditioning. elevated Consistently, showed stronger capacity. Besides, exhibit markers than induction. Moreover, reduced promoted deposits, when culture. Mechanically, induced increased reactive oxidative species (ROS) level activation ERK1/2, which closely related enhanced deposits better transplantation, preservation deposits. Conclusion These results suggest that medium boosts under normal or hostile culture condition via ROS/ERK axis. It promising pre-treatment promote regenerative capacity, superior untreated transplantation.

Mesenchymal Stem Cell in the Intervertebral Disc
Degeneration of the intervertebral disc (IVD) is a major spinal disorder that causes back pain. Nucleus pulposus (NP) in the central of IVD dehydrates and become more fibrous in the IVD degeneration. NP cells undergo apoptosis with the degeneration of extracellular matrix (ECM) components. To replenish the NP cells and core ECM, bone marrow mesenchymal stromal cells (BMSCs) have been highlighte...

Low back pain is one of the largest health problems in the Western world today, and intervertebral disc degeneration has been identified as a main cause. Currently, treatments are symptomatic, but cell-based tissue engineering methods are realistic alternatives for tissue regeneration. However, the major problem for these strategies is the generation of a suitable population of cells. Adult bon...

Degeneration of the intervertebral disc (IVD) is a progressive and chronic process, and the high incidence of discogenic disorders calls for new therapeutic approaches, such as cell-based therapies using three dimensional cultures and mesenchymal stem cells (MSC), which can differentiate to chondrogenic- and IVD-lineages. Here, we investigated the growth and differentiation of human MSC culture...
متن کاملHypoxia Preconditioning Promotes Survival And Clonogenic Capacity Of Human Umbilical Cord Blood Mesenchymal Stem Cells
Background: In recent decade, human umbilical cord blood derived mesenchymal stem cells (hUCB-MSCs) provide enormous potential for appropriate cell therapy, but they have limited growth potential and cease to proliferate due to cellular senescence, so providing a strategy for increasing the stem cell survival is necessary. Methods: In this investigation, MSCs characterized by flow cytome...
